I am trying to grow a single HEK293 cell in each well of 96 well plate.
It is been a week i do not see anything. does anyone have any experience?

you should see cell confluency between 50% and 70% of cell growth after 1 week ,Usually . you can replace your culture media by ATCC-formulated Eagle's Minimum Essential Medium +20 % FBS or RPMI 1640 + 20% FBS , 100 ul in each well . please ensure high viability and healthy cells before single cell cloning

It depends on the status of HEK293 cells you implant into the well of 96-well plate. Usually, cells in logarithmic growth phase are detached from bottom of culture dish or flask and then inoculated into each well at at the density of 10 000 cells per well，under this condition， it will take 7 hours at least for the suspended cells to attach to the bottom of the well and begin to grow. In the next day, change fresh media for the cells, at this time you should can observe the cells in growth under inverted microscope. According to your description, it seems that you lose the cells carelessly and no cell were successfully implanted at all. Or you can not use the microscope correctly. You should be guided by an experienced advisor before your own operation. Good luck!
