I am currently doing an MTT assay with different compounds like ATP, Adenosine and VPA. And I need to treat the cells for three days. Can I use the compound stocks made on the first day for the next three days also, or should I make freshly for all the three days. And I also want to know which chemicals can be used as a positive control and which can kill all the cells?
Hello Sai,
The answer lies in the activity and stability of the compound and it varies from company to company. Generally you can reuse a stock if it is stored very well at -20 and there is no need to make a fresh stock everytime.
As a compound which can kill all the cells, you can use 10-20% ethanol which can induce cell death very well.

I would prepare the stock and adjust the pH in advance. Then you can aliquot and store at -20 . At each respective experiment day, simply use one aliquot.
Thank you Mr. Ayata! I think I should do that. Will there not be any change in their activity if i store them in -20?
