how long the serum samples can be stored at -80C? i have intended to use them for the detection of cytokines (IL10) through ELIZA (commercially available).
It really depends on what information you plan to get from your samples. Many things can be tested for many years after collection when stored at -80C. But I agree with all above, analyze as soon as possible for your target.
I agree and we are currently using samples older than 10 years. This may be essential for research with human samples from individuals with rare diseases. An important thing is to keep the samples in criotubes to avoid freez-drying. Even though research freezers are not non-frost they are drying over time and concentrations of the target molecules may be increasing. I We always run and compare fresh and old control samples to estimate how good they are.
It is also important to be extremely careful when organizing samples or kleeining the freezer, keeping samples on dry ice when out of the freezer even for short periods of time.
up to our simple knowledge, this depend on what is the purpose for use of this sample.. in forensic sciences for specimens of human source like bone, teeth for for detection of mitochnodrial DNA, the extracted DNA Will remain for ever at which no degradation of mtDNA Will occur in contrast of nuclear DNA that will degraded easily.. for serum sample, storage in deep freezer will create favourable condition to remain active for few years.. A lyophilizer is long term condition if storage that will avoid freezing and thawing for sampke which cause a mutation in gene for samples for bacteriological and molecular studies..
If your serum sample is stored in small aliquots and the vial is properly sealed (parafilm paper) and preserved with sodium azide, it can last several years at -80 C.
My experience goes more along JR Vielma: If appropriately sealed to avoid dessication and treated aseptically or with sodium azide , and conveniently aliquoted to avoid freezing-thawing cycles you can get reliable results from biomarker detection for many years.
