I am trying to make gene deletion using FRT-mediated recombination in Drosophila. There are two p-element insertion with FRT sites across one particular gene. The distance between these two FRT site is 7Kb, I am wondering about whether or not these two FRT could work?
Considering the distance, 7kb, it should work - if you have the correct FRTs (compatible FRTs with the same orientation & screening method, e.g. see papers below), according to published data and own experience.
Parks et al., 2004: "... Supplementary Table 2 online provides a list of 519 deletion stocks, covering approximately 56% of CG isoforms in the genome (see Table 2b), which have been transferred to the Bloomington Drosophila Stock Center. These deletions average 140 kb in length (ranging from 4.5 to 659 kb) and delete an average of 25 CG isoforms (ranging from 1 to 83). ..."
Ryder et al., 2007: "... Each deletion uncovers an average of 44 genes or 368 kb. The DrosDel deletions are, on average, 2.6 times larger than those in the collection produced by Parks et al. (2004) ..."
There might be other reasons why it might not work, e.g. haplo insufficiency. Good luck!
If you want to determine if your region is haplo insufficient look at the classic paper by Lindsley and Sandler et al 1972 Genetics called "Segmental aneuploidy and the genetic gross structure of the Drosophila genome" appendix tables 1 and 2.
