I have two tubes with lyophilized primers. My boss forgot about them and found after 3 years at -20C. Is it worth checking them or better order new ones?
They would even work, if they were dissolved stock concentrations kept at -20°C. I used primer stocks which where much older than this without problems.
I agree with Christian above; they should work even if they were stored dissolved in water. I routinely use 2 and 3 year old primer stocks stored in the freezer of a domestic frost-free refrigerator without problems, and have occasionally used much older stocks (nothing fancy: conventional PCR, QuikChange) successfully.
Shelf life of printers on the bench is 6+ months. DNA is extremely stable, Even if done of the oligionucleotides have degraded there should still be plenty remaining for PCR top take place.
I agree with the above collegues. I also have tried disolved primers in water after 3-4 years and they worked fine. A single PCR will solve the mystery...
Don't worry dear. It will work. Just cheer and do your experiments with it. I m 100% sure. I have also experienced the same as Panagoitis has. Make sure there was not a single hint of moisture inside the lyophilised primer vials before diluting it.
