Lactic Acid Bacteria (LAB) have some unique characteristics, e.g, being gram-positive, catalase-negative, anaerobic, halo surrounding. But how to confirm that any bacteria isolated from any specific source are LAB? Having the above characteristics are not enough I think. There are some papers (not in the top journals) which identified the isolated bacteria as LAB just because their origin were milk or milk products, were grown in MRS media, and have the above properties!
So, if I want to confirm whether any bacteria are LAB, what should be the procedure?

You can only be sure with 16S rRNA gene sequencing. I also checked them for oxidase activity and motility (which both should be negative). The majority of LAB don't grow on MRS.

Identification of Lactic Acid Bacteria can use MRS-CaCO3 agar after grow in this media . You use catalase test and Gram dye then go to 16S rRNA gene or 16S rDNA gene.

So, all gram +ve, catalase -ve bacteria producing clear zone on MRS-CaCO3 agar are LAB?

That means, all LAB belong to some specific genera and those genera do not include bacteria other than LAB?
Or, in other words, do all LAB have a same specific sequence on the 16S rRNA gene?
I am not familiar with culturing LAB. Could you suggest appropriate culture conditions (such as how to create an ideal anaerobic environment etc.) for the isolation and growth of LAB, please? Any paper with detail procedure?
