So I am testing this material for bioburden, I immerse 10 g of the material in 200 mL of eluent and then filter through a 0.45 uM membrane 100 mL for a Total Aerobic Count and 100 mL for a Yeast and Mold count.
My question is this, because I'm getting confused from other opinions:
If each count will be reported as CFU per the 10g of material then, should I calculate the CFU multiplied by 2, as I only filtered 100 mL per each count from the 200 mL total I used to extract the microorganisms?
Hello, simple and easy answer yes you shuld multiply by 2. But if you perform analysis 1 filtration for each parameter without repetition you will not get reliable results.
Appears you sampled 5 grams (100 ml of the 200 ml total volume containing 10 gram material) for total aerobic count and an additional 5 gram (the other 100 ml) for yeast and mold. Each is cfu/5 grams. so yes - to arrive at cfu/10g multiple by 2. Terminology - suggest diluent rather than eluent and dispersion or dissolution rather than immersion.

Thanks to everyone for their answers. It is a textile material and I'm still experimenting for the best way to perform this analysis, I've performed it in duplicate for more reliable counts, and as it is a textile I'm getting a lot of lint that clogs the membrane. I'm thinking of pre-filtering on autoclaved Whatman filter paper grade 4 maybe for the Yeast and Mold count but I don't know if it would still retain a significant amount of fungal structures.
