Today i received a tetM primers forward and reverse and this is the first time for me to make a real time PCR reaction and i recived stock with 100(micro molar) that i need 218(micro liter) to get it ( according the Data sheet).
i want to prepare 900nm for one well, i have to take 0.45(micro molar) for one well..

What is your final volume per well? And What exactly do you want to do? Do you want to learn how you can prepare your primer? or how you can prepare RT PCR reaction? Depends on your answers, I will try to explain, if I know the answer.
