I have a question about protein-protein interaction. I am studying interaction between 2 proteins: RUNX1 and CBFb. In certain types of leukemia, there is inversion (inv: 16) for CBFb, which results in CBFB-MYH11. What is published is that CBFB-MYH11 has more affinity to RUNX1. If I do an immunoprecipitation I can see interaction between RUNX1 and CBFb as well as RUNX1 and CBFB-MYH11. Can anybody suggest an experiment in which I Can check how much % of RUNX1 is bound to CBFb and how much % to CBFB-MYH11? One simple experiment came to my mind is to do immunoprecipitation with CBFb and MYH11 separately and blot for RUNX1 and quantify RUNX1/CBFB and RUNX1/ CBFB-MYH11. But I think this will not be quantitative. Please let me know your thoughts.
