RESPECTED FELLOWS KINDLY HELP ME.
I am developing a nanoparticles formulation but it is continuous precipitating . Ingredients used in niosomes are TWEEN-80 , SPAN-20 , CHOLESTEROL and buffer solution 7.4 pH . My optimized formulation is now precipitating in the evaporatory flask immediately as i hydrate it. i check out multiple parameters such as solubility, temperature, ingredient source , quantity , buffer etc but i am unable to find the exact cause of that precipitation . kindly guide me if possible please

One way to overcome precipitation is to induce charge (zeta potential) to a quantity + / - 30mV in your vesicles.

Are you at liberty to say what API you are trying to encapsulate? Is this phenomena happening with empty niosomes? How much cholesterol are you adding?
Respected sir , I am facing same phenomenon of precipitating with out API . Cholesterol is 50% of total formulation ingredients sir
Precipitation is a matter of density of particles which result in sedimentation due to gravitational forces. As I mentioned above,
try to create repulsive forces (charge) between your particles by using charged ingredients (e.g. DCP);
and:
check the densities of your niosomes and the buffer in which they are dispersed in ... simply, in order to have something hanged in a suspension medium, it should have same density as the medium ...
It would be important to assess the contents of the precipitates. In addition, use the suggestions of Professor Mozafari. It could be anything from as easy as the volume of hydration medium, it's pH, or even the total cholesterol in the formulation. Knowing what is actually precipitating out could be cardinal. The HLB of the surfactant mixture is also cardinal.

What is the hydration medium composition? Did the API is added in the hydration medium or it is added along with tween-80 and span-80. What is the solubility of the API in the hydration medium? As mentioned by Bwalya Witika it may be due to a high amount of cholesterol. Please share with us the step-by-step procedure along with details so that we can find a solution to your problem.
