I prepare the solution of curcumin with 50% ethanol and 50% water. I want to investigate cytotoxcity by MTT Assay on cell line MCF7. but the problem is cytotoxcity of ethanol. how I can solve this problem?

You can try to prepare a concentrated stock solution of your sample and then dilute it in the cell medium to reach a final ethanol concentration below 0.1%

i agree with paolo , you can do :
1- curcumin Soluble in ethanol (10 mg/ml), DMSO (25 mg/ml) as stock solution
2-you can used "AQUACUMIN" , aquacuminis the new innovation for the curcuminoids that well soluble in water up to 150 mg./ml.

Hi Elham,
As Paolo and Reza already suggested, you may also use DMSO for concentrated stock preparation and can further dilute it with media to reduce toxicity.
It is also important to have a control group with the same concentration of EtOH/ DMSO but without curcumin in your experiments.
Ethanol can act as a phytoestrogen and MCF7s are ER+. So use DMSO instead, 0.1% (v/v) and have 'DMSO only' wells to normalise to, as suggested above.
Richard Elliott is pointing to an important issue of avoiding the use of ethanol to solubilize curcumin as ethanol could influence the result of your experiment. Please use DMSO as recommended by Richard.
