Most of the researchers use Iodine solution for screening of Pectinolytic organisms. When pectin is used as the sole source of carbon and when an organism is grown on that medium, it would hydrolyze pectin if its pectinolytic in nature. As a result the iodine would not be able to form complex with the pectin and that area would remain transparent whereas the area wherein pectin is present a yellowish brown color would develop. From the transparent zone you can easily idenitfy whether the said culture is having pectinase activity or not. Good lick for your research....
Unless and otherwise there is a contamination of polymeric substrates other than pectin the chances of false positive or negative reactions is very very low.
