I want to isolate fibrolytic bacteria from a sample enriched with plant material but I'm assuming that they will be attached to the plant fiber instead of being in the supernatant after making a suspension and centrifugation. Are there special techniques or reagents that need to be added to dislodge these bacteria? Or is centrifugation, in fact, enough?
A combination of chemical and physical methods should detach adhering microorganisms from solid surfaces by a high percentage. Although we do not have experience with plant fibers, we got good results when using sodium pyrophosphate and sonication to detach organisms from soil and sediment particles according to the method of Amalfitano and Fazi (2008) Journal of Microbiological Methods 75:237-243
Hi Jeremy Martin Torres, i would like to know if you have tried the recommendation of Helmut Brandl or if you tried/found another solution. I have the same problematic, in my case, i would like to detach the microorganisms (bacteria and fungi) from flax fibre.
