I have been having trouble identifying a good range of doses to use for a drug synergy matrix.
So far, I have tried
1. using 0.1, 0.3, 1, 3 - like increments with the IC50 value on the 7th value out of 9 values.
2. doing 1:10 dilutions with the IC50 value on the 8th value out of 9 values.
The first time, I wasn't sure if synergy could happen at a lower dose than I have tested for.
The second time, no synergy appeared at such low doses and there was just synergy at the IC50 value. The individual dose-response curves were very vertical.
How do you set up your synergy matrices? I feel like I'm missing the sweet spot of values to use.
I had trouble setting the intervals and the maximum concentration in the matrix format, as I had many cell lines with different sensitivities to each drug.
The way I managed this was to set the highest concentration
1. lower than the Cmax value, or
2. at the highest concentration where we see an effect
for all cell lines tested.
I divided the concentrations into 5 values, since this is what fits on a 96 well plate with replicates. (ex: 0, 0.03, 0.3, 3, 6uM)
