I am looking to prepare an egg yolk supplement in-lab to support the growth of Mycobacterium avium subspecies paratuberculosis in 7H9 liquid culture. A method was presented in a paper by Harris et al 2005 (http://vdi.sagepub.com/content/17/6/554.long) but there is no mention of sterilization or how to aseptically separate the yolk from the albumin. Has anyone ever prepared the supplement in lab before?

According to a former microbiological colleague it is quite simple.
Take fresh eggs as the yolk membrane decreases in strength over time. You may treat the outside with 70 % ethanol.
Then aseptically break the egg and separate egg yolk and white by rolling the yolk over a sterile surface until all the white has been separated from the yolk. Take a sterile needle and collecting device and puncture the yolk membrane to collect sterile yolk material.
Will take some time to gain experience but then it is a piece of cake!
