In an experiment where you are testing the effects of two drugs on tumor burden, how do you account/normalize for changes to body weight? For example, if one drug results in slower weight gain in mice compared to the other, how can one normalize for that change to say that any effect seen on tumor burden is still apparent despite the changes to animal body weight?

How do you measure the tumor burden in your animals? Is it tumor size, weight or count of tumor foci?
I do not work with animal models, but simply dividing tumor burden per animal weight wouldn't work for your purposes?
Alternatively, regression models including both the drug and the animal weight as explanatory variables and tumor burden as the dependent variable could be useful to evaluate the "weight" of both variables in the outcome (tumor burden).

We measure tumor size throughout the treatment period and then both size and weight once the mouse has been sacrificed and the tumor has been removed.

I am not sure but I believe that expressing the drug dose on the basis of metabolic body size and studying the change in metabolic body size, instead of absolute body size (say, body weight) should serve your purpose.
