Have a look at the following protocol which is from our laboratory. We have 95% success rate.
Diabetes is induced in overnight-fasted rats by a single intravenous injection of STZ (50 mg/kg) freshly dissolved in cold citrate buffer (pH 4.5). Rats are tested for successful induction of diabetes, at 72 h after STZ induction, by measuring the fasting blood glucose level. Only those rats having blood glucose level >250 mg/dL are used in the study. If you have any query, let me know. Good Luck!
Yes. I agree with Bhesh. But STZ at 65 mg/kg by intraperitoneal will also do. Beyond the protocol induction depends on the quality of STZ you use. Try to avail from sigma and that will work.
I usually induce diabetes in rats by a single intra-peritoneal
injection of streptozotocin (60 mg/kg). Glucose blood levels are measured in 2-hour-fasting animals 2 days after STZ injection. Two days after STZ administration, glycemia rises (approximately 500/600 mg/dl) and this effect is maintained for a long time.
