I want to study the release of doxorubicin using dialysis. The problem is that while doing dialysis, the membrane absorbs most of the drug and actual gradient causing the diffusion of drug to get disturbed of which I am not getting a proper release profile.
I am using deionised water to dissolve my drug and for dialysis, too. Does it have something to do with the pH or the membrane? I am going to try PBS for this release to check if it works, but please help if you have faced a similar problem.
I dont think it absorbs. There may be problem with molecular cutoff problem. Better take 10k mol wt cut off membrane
Hello, I'm going to do the same experiment to have it as a control, I have a question, use doxorubicin powder and placed in the dialysis bag, or adds a certain amount of solvent in the dialysis bag with doxorubicin?
Hi Binh Pham , I had to use 3.5 kDa only as I was working with graphene quantum dots, which would have come out along with doxorubicin if 10kDa was used.
Thank you for your respond. So the size of column was not a problem. In my case, I tried to change the solution disolving my drug (deionised water, PBS), and increased the amount of drug in dialysis membrane but it was not really effective. Could you have any recomendation for me?
