Which plasmid should I consider for quantification using DNA ladder as concentration standard? Plasmid has to be used for restriction digestion. I need an alternate to spectrophotometric method.
Thanks. My concern is, which plasmid confirmation do I quantify?
Your question is not clear. Do you mean which band do you look at? Or are you asking what plasmid you should use to create a DNA ladder?
I need to know if there's a way I can specifically quantify (visually) his-tag fusion protein. I have checked for commercial his-tag stain reagents, but I'm looking for state-of-art method(s)....

I am looking for cheaper and still efficient state of art methods. I have checked for FACS and a few commercial kits but they are out of my budget.

In their work (see attachment), Pansri et. al. has used pull-through PCR. I am unable to understand the purpose because I do not know about Pull-through PCR.
