Even I have the same question. I am in need of DNA from Zoanthids. I tried one method of DNA extraction from Sea anemones, from which I was successful in first attempt. You may try that method.
hi try this, Samples preserved in ethanol were rehydrated in sterile distilled water for 1 h. About 50 mg of tissue was taken and add 1 ml of guanidine hydrochloride buffer (10 mM Tris HCl, pH 8.0, 0.1 M EDTA, pH 8.0, 6 M guanidine hydrochloride and 0.1 M sodium acetate) and homogenized well,Then the homogenate was transferred to a 1.5 ml microfuge tube allowed to react for 30 min at room temp, then centrifuged at 5000rpm for 5 min at 4°C. Supernatant was transferred to a fresh microfuge tube and add equal amount of 100% ethanol centrifuged at 14,000rpm for 20 min at 4°C, discard the supernatant then add 95% ethanol centrifuged at 14,000rpm for 10 min at 4°C,discard the supernatant then add 70% ethanol centrifuged at 14,000rpm for 10 min at 4°C,discard the supernatant,Then the DNA pellet was dried in a vacuum drier and dissolved in 50 μl sterile distilled water. A portion of this extract was used for
The best DNA isolation kit is Qiagen for Marine Species. You can also DNA isolate from Salt and Phenol chloroform, before that you have to check two important point.
In your case Ethanol preservation will may create some problem, check the quality of ethanol, try to use Analytical grade ethanol, and before store in Ethanol, you kindly check the quality of shrimp specimen, if it is started to degrade or spoiled one you avoid that one for store
Which manual method is successful for u Purushotaman. When compared to qiagen and manual method which is the better one wrt to down stream application like PCR, qRT PCR, Sequencing?
