Hello! I am having problem in sorting BMDCs from a heterogenous population of murine hematopoietic stem cell which have been cultured in RPMI+10% FBS+ 1% PenStrep with FLT3/L (for differentiation) for 9 days. Every time I have sorted the BMDCs, either the cell morphology completely changes or the cells die after a day. Any help regarding this will be appreciated. Thank you.

Thank you for your answer. I use 1xPBS with 10% FBS as sorting buffer. And I collect the cells in RPMI+10%FBS+1%PenStrep
You may be using a too high run speed, the nozzle size may not be suitable for DCs, pH and temperature of the collection solution may need some adjustments. I recommend to contact the product specialist of the company you have purchased the sorter from. They will help you to adjust the flow rate, pH and most suitable combination of ingredients for the collection solution.
