I am doing primary culture of cancer cells, but I am facing fungus contamination. Does anyone have any idea what should I do. I really appreciate your help and time.
Use 1% pen/strep and 0.5% Amphotericin B in the cell culture media. Throw out all the cell culture media which may have fungal infection. Resterilise your pipette tips and autoclave PBS or any DIH2O used. Clean all your incubators with ethanol and virkon and clean out the laminar flow units from top to bottom with warm soapy water containing Virkon. Also, if you have another batch of cells it is best to dump any infected ones as fungus can spread rapidly in a tissue culture environment. You can also test specifically for remaining fungal infections in incubators by placing a small nutrient agar petri dish on the bottom shelf. I hope that helps!
