For now, I am still planning to do the following:
1. Inoculate isolated colony in 50 mL TSB medium
2. Incubate the culture overnight at 27 degrees C
3. Inoculate 1 mL of overnight culture into 10 mL artificial seawater microcosm (with different pH, salinity and temperature)
4. Incubate microcosm at different temperature (4, 27)
5. Check the CFU every 24 hours by spread plate on Trypticase Soy Agar until stationary or death phase
