I am in the process of learning to calculate IC50 values of OCI AML2 cells for Cytarabine. I have taken the drug concentrations as 0.01 uM to 50uM. My question is while calculating IC50 is it essential to convert my drug concentrations from micromolar to molar and then take the log values for graph plotting or directly go without converting into their molar values.
It is not necessary to convert the drug concentrations to molar. If you use the micromolar concentrations, the IC50 measured will also have micromolar units.
It is not necessary to convert units in your calculations because IC50 is measured in the same units. IC50 is the point on your diagram where you have 50% of inhibition for specific concentration of used inhibitor.
Hello The conventional way to calculate the IC50 is by plotting the log [M] on the x-axis. Software like GraphPad Prism requires it to display the result of the IC50 calculated in [M], where if you get an exponent "10^-6" you will immediately know what it is [uM]. The essence of graphing them like this is to obtain a standardized and readable chart for any researcher.
