hello. i manually measured the area in each image. the output i got was area/mean/min/max
then i put all the data into excel, and also added the following columns: group names/date/hour pic was taken
i have several drug groups and 2 control groups (DMSO and untreat) - however i only want to compare them to DMSO. untreat would be nice but not necessary
my question is: how do i analyse the data? what test should i use and in what way? i would be very grateful for any tips!

Although I can't answer your question, I have one regarding the conduct of your experience; Can you tell me how did your perform your scratch ?
Because, i'm realizing this test using a P1000 tips and I guess the cells i'm using are may to aggressive and close the breach too fast.
Could your recommand me a way to do a larger one ?

In our experiments, we used pipette tips of 0.5 - 10 microlitre mostly in 24 well plates . and we took pictures at 0,24 and 48 hours. I can't comment on using a 100-1000 microlitre pipette tip as we never used that. but from my experience, if you put more pressure on the tip when you do the scratch, the resulting scratch will become bigger . personally, nothing comes to mind that would be useful for making an even bigger scratch than that . maybe in the worst case scenario, take a cotton swab, remove the cotton, keep the plastic, autoclav it and use that? or some similar object that can be autoclaved.
