I generated iPS cells from urine cells of tumor patients. I want to induce tumor from iPS cells in vitro.I did specific differentiation and plated differentiated cells onto soft agar,but failed. How could I do? Could you give me some suggestion?
Did you have any indication that plating of differentiated cells onto soft agar would yield tumors? Have you seen this before or found a reference indicating that it would work?
Soft agar is used to test whether your cell line is tumorogenic. Not every cell line is going to just form a tumor. You could try hitting your cells with a known mutagen, or perhaps try a small molecule agonist or inhibitor for a tumor suppressor/growth protein (crazy topic I want to try myself).
