HEK293/DDR2 showed a little increase in ROS production in comparison to HEK293 cell line, but ROS generation was found to be decreasing in HT1080 (HA-DDR2) cell lines in contrast to HT1080 cell line. Its puzzling to have such conflicting data.
Looking forward to have suggestions.

First you need to define ROS production. what is the reporter? how do you measure it? how do you express ROS units? how do you normalize the signal?

We have been doing the DCFDA assay, by measuring the "end point" with a multiplate reader. I am not sure about he last question of yours. I'll be much obliged if you could shade some light on that.

Normally in cell lines could get different clones derived from few genetics differences amongst cells Also ROS production or tolerance to them could be depended to the cell cycle state. I suggest treat cells with cis platinun or H2O2 or other drug which triggers ROS production and studied if you have a subpopulation that survive to the continuos treatment with this drug , then tested in these if you could found the decider effect.
