Roland, Are you looking for a way to mark injections during an experiment to verify intrathecal placement or are you trying to find a way to verify placement during training of the procedure?
Both if possible. Both color and morphine can be used to verify correct IT positioning during training. For experimental purpose, as animals will be kept for longer period following IT admin, I don't know wether this is feasable to mark IT spot and to be visible several weeks later. roland
Unfortunately, in my experience, I have been unsuccessful in finding an agent that can be used during injection to later (sometimes weeks) positively confirm location of administration that does not wash out or have some effect in and of itself during administration. In my experience, we have used injection of bombesin during training until trainees can consistently get 95% positive bombesin response before they can move on to actual experimental studies. The injection causes an immediate local itch response at the flank that lasts for a few hours. Lidocaine and morphine can also be used in this manner. In instances using a cannula, you can simply inject dye at the time of sacrifice to verify placement but to my knowledge there is no surefire way to verify every single placement during an experiment without introducing another variable or changing the timeline to accommodate possible washout, etc.
