I want to know how we can precisely trace the lineage of cancer progression. Is it originate from the mutation of a Normal cell or a Stem cell? Is there any method available to precisely delineate the origin, even if we consider the heterogeneity of cancer cells?

Cancer cells always originate from a mutation of a normal cell. Usually more than one mutation is necessary. e.g. retinoblastoma originates through two mutations: one in each alele of Rb gene. Two hit of Knudson.
Colon cancer requires at least five or six different genes to mutate.
It has not been proved but it is possible that epigenetic modifications may produce cancer. e.g. methylation of Pten gene in prostate cancer. This requires further testing.
Independently of it is a mutation or an epigenetic change there is always a genetic cause in the origin of cancer.
