I am currently conducting an experiment in which I aim to induce diabetic ketoacidosis (DKA) in rats in order to measure isopropanol levels. While I have been able to successfully induce diabetes through streptozotocin injection, I have encountered difficulties in consistently achieving a clear state of DKA in the animals.
I would be truly grateful if you could share any advice or references.
If the classical streptozotocin (STZ) method isn't working, there are alternative approaches to induce diabetic ketoacidosis (DKA) in rats. Here are some options:
- Alloxan Induction: Similar to STZ, alloxan is another chemical that selectively destroys pancreatic beta cells, leading to insulin deficiency and hyperglycemia.
- High-Fat, Low-Carbohydrate Diet: Some studies suggest that feeding rats a ketogenic diet can push them into a diabetic state, especially when combined with insulin resistance models.
- Genetic Models: Certain rat strains, such as Zucker diabetic fatty (ZDF) rats, naturally develop diabetes and can be used for DKA studies.
- Hormonal Manipulation: Administering glucocorticoids or other hormones that affect insulin sensitivity may help induce DKA.
- Combination Approaches: Some researchers use multiple low-dose STZ injections instead of a single high dose to gradually induce diabetes and ketoacidosis.
