I was wondering how can we experimentally figure out the presence of any given gene in a chromosome in the human genome. For example we know that the gene for alpha- Amylase is present in chomosome 1, but how to experimentally prove it ?
Design a primer specifically to one of the exons of the gene and amplify. To know if it is the exon of the particular gene that you are looking into sequence the exon and match it with the gene.
Numerous techniques were used before the framework human genome sequence was available, in fact many methods were used in construction of the framework.
A few of the methods are FISH of metaphase chromosomes, fiber-FISH, and radiation hybrids. Depending on the precise question and how the methods are implemented, they can prove that a sequence is on a chromosome and the physical proximity of other sequences known to be on the chromosome (high-resolution mapping).
