PLGA nanoparticles (200 nm) showed an extravascular-like plasma profile after intravenous administration with peak concentration after 5 h. Then upon conjugation to cRGDyk (which showed no change in size but decreased zeta potential significantly “became less negative”), normal intravenous profile was witnessed and peak concentration was observed 15 min after administration.
Also, hybrid lipid-PLGA nanoparticles (200 nm) showed a similar extravascular-like profile but upon peptide conjugation (same size but lower zeta potential), the elimination phase disappeared and a continuous increase trend was observed up to 24 h post intravenous administration
This experiment was carried out in melanoma-bearing C57 black 6 mice.
