I need to prepare L-tyrosine to prepare some modified M9 medium but while preparing it in water it remained turbid and adding HCl while stirring makes it clear but at the same time HCl lowers it pH (required pH of L-tyrosine is 7.0). I need to filter sterilize it after it get dissolved in water. Can you suggest something on this?
Depending on your desired final concentration and whether or not you are using liquid or semisolid media it might be possible to dissolve the L-Tyr in your media then filter sterilize the aqueous solution instead of autoclaving the media and adding the filtered L-Tyr.
Alternatively, as the solubility of L-Tyr given in the attached Sigma product sheet is .45mg/ml in H2O the L-Tyr could be dissolved in H2O at a low concentration. E.g. 112.5 mg L- Tyr / 250 ml H20. this dilute solution could then be filtered and added to the media with the 250 ml H2O being accounted for in the final volume. This could present an issue depending on the media components as the reduced volume could in some cases result in precipitation.
You should use progressive dilution as above or try 2% ethyl alcool in the stock solution
Dissolve the L-tyrosine in a small amount of organic solvent (eg. DMSO) then dilute the solution in water to the desired stock concentration. Add the L-tyrosine / DMSO mixture to water drop-by-drop and under continuous agitation.

I have tried similar methods to that of Mrs. Barbulescu with limited success. I found the use of DMSO to be somewhat ineffective with both L-DOPA and tyrosine. This is the reason I suggested the method above utilizing an aqueous solution. I found that the DMSO and L-DOPA or Tyrosine would both drop out of solution when added back to an aqueous solution.
I also tried this DMSO method but it didn't work, it is falling down.
Suggest me something i need to make 10 mM solution of tyrosine in water
