I have prepared 0.8% agarose solution in 50mM calcium chloride buffer. 2ml of 5% SDS solution was added to the agarose solution as surfactant and then, 2 ml of FluoSpheres™ Carboxylate-Modified Microspheres were also added while mixing the solution continuously. I was expecting these fluorospheres be stabilized in agarose solution. However, the confocal imaging shows the agglomeration of the nanoparticles. When I changed the calcium chloride buffer to PBS, ph 7.4, the particles seem to interacting with buffer. I have attached the imaging pictures.

Does anyone know what actually went wrong?
