Freeze dried bacterial cultures maintain stability even when stored at ambien temperatures for a long time. This applies to cultures that have been dried to attain a certain moisture level usually determinable during the process by some advanced freeze dryers. However for most old freeze dryers one has to have a different way or an indirect way that does not compromise the integrity of the sample.
Three major methods are reported for the determination of moisture content or residual moisture of freeze-dried products. These include Titrimetric method, (Karl Fischer), Azeotropic method, Gravimetric method (Loss-on-drying). The gold standard method in current use is Near Infrared spectroscopy.
