My cultures are grown on tryptone soya broth (TSB) and nutrient agar slants. Those on broth are centrifuged and pellet suspended in the same growth broth with equal volume of 20% glycerol before dispensing 1.5 ml of cell suspensions in to vials for freezing. Those on agar are washed off with 10% skim milk and equal amount of 20% glycerol added before dispensing 1.5 ml of the suspensions in to vials for freezing. After freezing for 3 hours at -50 degrees Celsius the vials are transferred in to an acrylic drying chamber, when the condenser chamber is around -50 degrees Celsius and vacuum applied. In the first trial some of the suspensions completely lost while others melted before the freeze drying was over. In the last trial most vials lost significant amount of the solution while others retain a small amount of a jelly like deposit of the suspensions.
