I am exploring the function of one peptide drug in different cell lines. I didn't use cell culture medium like DMEM to dilute the peptide because I thinks FBS in the complete medium might degrade peptide. Thus, I plan to dilute the peptide into opti-MEM then treat cells. But I have no idea how long the cells can tolerate with the low-serum medium. Additionally, as it was reported that low-serum condition might change some protein expression which might affect my further experiment of protein detection. Can anyone give me any suggestions?
Actually, fetal bovine serum is considered to be inhibitory of proteases. So your peptide would most likely be safe from proteolysis in your growth medium containing FBS.
