will it be more reliable to transfect the cells during culture or transfect after exosome isolation?
Dear Eman Thabet
It will be more reliable after exosome isolation.
During exosome transfection with miR inhibitor, you have to ensure following,
1)quality of exosome,
2)Need to standardize the concentration of exosome localize to cells.
3)Need to standardize also a concentration of miR inhibitor
4)While transfecting exosome makes ensure miR inhibitor encapsulated into exosome.
4) List of methods available for transfection such as
Chemical based: Lipid, Calcium phosphate, cationic polymers, DEAE-dextran, activated dendrimers, and magnetic beads.
Instrument based: electroporation, Biolistic, microinjection,laserfection/optofection.
Virus-based methods. Do literature collection and select which method mostly used for publication and then adopt that one.
Sincerely
Prabu
what is the best way to measure exosomal CD markers(such as cd81,63,tsg101) ? are western blot results reliable? what are the challenges?
