I was using Triton x-100 before, but when we start using the pLysS bacterias it was imposible to keep using it. What we are trying to do is to stain inclusion bodies with an amyloid protein but we need to permeate the cell to be able to stain it with Thioflavin S. Apparently the cell membrane is not capable of supporting this type of detergent , i also tried to do the same with Tween but I have the same results. I would like to know if there is any way to permeabilize the cells other than using those permeabilizing agents and that allows me to have reproducible results because we think that maybe using some parts of the bacteria trasformation protocol would be usefull but it is not reprofucible.
