I want to know how to detect activation of PKC alpha. Does an increase in the phosphorylation level of PKC alpha corelate with the activation of PKC alpha? Please explain.
I am not an expert with PKC, but it seems to have a usual activation loop phosphorylation site like many other kinases (aa 400+), required for full activation. In addition a C-terminal site (T638) which is an autophosphorylation site. The first one will be required for activity, while the second one should be real read out of activity. I would generally assume that if there is autophosphorylation, the kinase is active. Hope it helps.
As manoj mentioned, phosphorylation levels on threonine 638 of PKC alpha, an indicator of PKC alpha activation (Bornancin and Parker, 1996; Parekh et al., 2000).
