How can I screen natural products/compounds for AMPK activity in 3T3-L1 cells?

I plan to use ELISA kit to measure AMPK activity. This is for screening natural products for potential activators. Should I measure both total and phospho ampk or phospho ampk alone. Can anyone suggest the best kits for the same.

-how pure is the your natural products?

-water fraction or organic fraction

-can the natural product to passing to the cell memebrane ?

-polarity, and stability in biological
