Currently we are performing transfections of AAV-2 plasmid DNA from Cell Biolabs AAV2 helper free plasmid packaging system into HEK293T cells. This is serotype 2. We would like to change to serotype 8. The information that I have been given is that we only need to change the rep-cap plasmid and can still use the helper and GFP expression plasmid from this kit. Do you have the a repcap serotype 8 plasmid? If not do you have any suggestions as to what other companies I should contact.
Hi Paula, you just need to replace the RC2 plasmid to RC8, a AAV8 would be produced when you transfect 293T cells with 3 plasmids: AAV vector, RC8 and pHelper.
