Anything with free amines can be used theoretically. Tris-HCl buffer, Ammonium chloride, and ethanolamine are commonly available good options.

If chemicals may be harmful for your cells, thus better not using them. You should wash extensively by refluxing hot water in a Soxhlet extractor. It is very efficient for throroughly washing materials during several days. As the system is in closed loop, no water is lost so that you can wash as long as you want with the pure water distilled from the system itself.

reading your comment to your question, I suggest to use e. g. 50 mM NH4Cl (Ammoniumchloride) in buffer solution. This is used also for free aldehydes after fixation for Histochemistry, IHC, EM etc.

My typical routine steps are (after the fixation)

NB: use a if available

1) washing in appropriate buffer solution (e.g., the type you used with the fixative for at least 5 min @RT (depends naturally also on the size of your specimen), just to get rid of most of the fixative (GA-solution) proper. Decant the washing solution.

2) incubate at least 1 x 35-40 min @RT specimens in

washing buffer as in 1) + 50mM (or 0.5 M) NH4Cl (ammoniumchloride, you'll find that it will dissolve rapidly on shaking...you can make stock volume, e. g. 1 L, don't forget to label your flask/bottle unanimously!) .The incubation time in the original recipe (see below ROTH et al, 1981) has been up to 4 hours @RT, followed by (cf. also step 3 below) pure buffer solution proper over night at 4°C.

3) again incubate in washing buffer as in 1), at least 1-2x for 5-10 min (depending on size of your specs).

When I looked through the literature, I saw that people use glutaraldehyde as a fixative agent before staining or microscopic assays as you mentioned. But I could not be sure that I was on the right way or not. With your explanation I understood better. Thank you very much.

NaBH4 will reduce the glutaraldehyde into a diol (which is soluble in water). If you have LiAlH4 in your lab. ,then try it and it is even more effective reducing agent. Once you get the diol, the answers given by the scholars to you offer good ways to wash off the chemical from the cells.

Thank you for your answer. I found ammonium chloride and glycine in my lab. I am not sure if we have LiAlH4. I am still wondering about which buffer (and pH) I should use to make a solution with them. For example if I use phosphate or Tris-HCl buffer to make solution, then I should use it as a washing buffer. Is it right?

Glycine is the most commonly used quencher, Tris works by itself as a quencher too. So you can prepare a glycine solution in Tris to achieve quenching. Such a buffer is already available commercially.

Tris-Glycine buffer is generally used in Western Blotting, I don't know if it has been used for quenching but you can give it a try. Theoretically it should work.

We found that GA detoxification of aortic wall tissue required extended warm washes with amino reagents, and low pH to depolymerize and extract hemiacetals that may have formed (depending on the quality and concentration of the GA used). Reduction of the imines and aldehydes with NaBH4 is also recommended.

Free glutaraldehyde is toxic for cells not the crosslinked one. So u have to remove uncrosslinked glutaraldehyde. U can remove it by water wash. Wash the sponges with distilled water several times and test the filterate for Benedict's test or Fehling's test. If the test gives negative result then sponges doesnot contain uncrosslinked gluteraldehyde

Free aldehyde groups can be blocked with either polysine (for immunohistochemistry) or any basic amino acid or addition of Tris buffer.

I was involved officially in preparation and realization of Works Council elections/polls for 4350 employees in our Hospital. Concerning your question to me (no 5 above) I would like to say that if you have "blocked", "quenched" or "deactivated" free aldehyde groups in your sponge samples you better should "wash" your samples at least twice for 3-5 min with a buffer (i.e. principally the fluid you dissolve the NH4Cl or any of the other possible chemicals as mentioned above). It can be phosphate or any else buffer, yes, distilled water also will be an option if your samples are not critical in terms of preservation of morphology or any other reason. If washing with A. dest you certainly will have to change some times (more than 3 times) and, as Anita G Sullad said, proof of lack of any free GA-molecule should be tested by Benedict's or Fehling's test). I have been using glutaraldehyde for short duration crosslinking of decellularised bovine pericardium. To neutralise residual glut, what I do is one hour wash with several changes of NS followed by 3 day treatment with 1% glutamic acid for aldehyde capping My material passes cytotoxicity test (both on extract as well as direct contact). The problem with glut is, after implantation in rat model, after several days it will degrade and generate free aldehyde resulting in persistent chronic inflammation. Dye mediated photooxidation, instead of glut crosslinking solves this issue.

Just a comment to get reproducible results in long lasting experiments. Reaction with primary amines works good in blocking free aldehyde however the reaction gives rise also to Schiff bases that might exhibit some degree of instability with time thus regenerating the free aldehyde group and hence its cytotoxicity. Therefore the use of NaBH4 would be advisable for stable and reliable blocking of such substrates.

Dialysis technique you can use 

I have been successfully able to leach out glutaraldehyde and other toxic reagents by giving a 48 hrs wash to the hydrogels. I let the gels stand in PBS for 48 hrs. Alternatively, you can incubate your samples in PBS for 24 hrs and keep them on a mechanical rocker.

maybe you can use HPLC to help remove glutaraldehyde residues. Protein has adsorption effection with some special columns, but glutaraldehyde doesn't have this characteristic, and you can isolate them in this way.

Glutaraldehyde is used as a biocide in medical, industrial, agricultural, oil and natural gas industry, and laboratory settings. Glutaraldehyde degrades rapidly under both aerobic and anaerobic conditions in water and under aerobic conditions in soil.

Glutaraldehyde is hydrophilic and can be removed from the atmosphere by wet deposition. Glutaraldehyde can be deactivated chemically by adding a suitable amount of alkalinating or reducing agents. Heat removal, carbon oxide treatment even water may be also one of major solution for the same. Two options are available, depending on the concentration of glutaraldehyde to be deactivated. With Sodium Bisulfite and With Sodium Hydroxide (Caustic Soda) could be used.

An effective chemical method that can be used to deactivate concentrations of glutaraldehyde (up to 5%) is by addition of sodium bisulfite (SBS). In order to assure rapid, complete inactivation, it is recommended that 2-3 parts (by weight) of SBS be added per part of active glutaraldehyde. Addition of 2-3 parts SBS will rapidly reduce the concentration of glutaraldehyde in solution to less than 2 ppm active within five minutes at room temperature.

Adding a sufficient amount of sodium hydroxide solution to maintain a pH of 12 will reduce the concentration of glutaraldehyde to approximately 20 ppm or less within 8 hours at room temperature. Maintaining the pH at 12 or higher is critical, as a lower pH will result in a significantly slower deactivation of glutaraldehyde. CO2 treatment and heat based removal of glutaraldehyde can be done. CO2 can be used to extract residual glutaraldehyde from crosslinked collagen (Collagen is the most abundant protein in the human body) films. Since CO2 is a nontoxic, nonflammable substance that is relatively inert and can be used to process biomaterials at mild pressures and physiologic temperatures. Collagen scaffolds require crosslinking to improve mechanical strength, requiring agents like glutaraldehyde that have high residual cytotoxicity. First, reduce glutaraldehyde concentration in ppm level or percentage. CO2 treatment can have negligible alteration of thermal stability but can significantly increase film stiffness and tensile strength. However, these changes were minor compared to heat-based removal of glutaraldehyde.
