Is there any isolation protocol for only cancer associated fibroblast (CAF)? According to the literature, there are some biomarker for CAF but it is not enough for characterization of CAF because of the tumor heterogenity. I want to evaluate tumor microenvironment without culturing the CAF.

You maybe able to remove CAF by using a FACs cell sorter after incubating with the appropriate antibody against the biomarkers providing the biomarker is expressed on the outer leaflet of the cell membrane.

CAFs are FAP+/CD90+/CD31-/CD45-/EPCAM-
As, Jack suggested, the best way will be to use flowcytometry. CD31 marks non-CAFs endothelial cells so you need at least these markers to be sure you are actually counting CAFs.
