I would like send DNA samples for sequencing to company outside my stay country for gene sequencing service. The company that will do the sequence service requested me to send the samples in room temperature; kindly supply me your experiences for preserving DNA samples in room temperature for minimum one week interval.

The company you sequence with should have a protocol for shipping DNA. Having said that, what I normally do is elute the DNA in TE and incubate at 80c for 20min to make sure any DNAse in the sol. is inactivated. Make sure to note the sequencing facility that the DNA is in TE when you ship it. I also sent plasmid DNA lyophilized, haven't tried it with genomic DNA but you can try lyophilizing it keeping it on the bench and pcri it.
Also if possible, try and expedite the shipment.

Firstly What Is The Source Which You Have Used For The Extraction ,
Is it A Crude Method or The KIT Based Extraction , Usually The The Stability Also Depends On The Lenght Of Your Mer Size and Apart From That Whether Any DNAse content Present or Not ?
And For Storing It Te Buffer Can be added and lyophilized under Ammonia Gas .
Upto 3 Weeks It Can Be Stored In RT .

Thank you for the answer. Have you tried before storing DNA in TE Buffer and Lyophilized under ammonia gas ?Does it good ? For more information the company request me to store it in Agar Plate/Glycerol Stock since the intend segment is 16sRNA , do you have any idea about this method ?

u can use DNA stabilizer such as Biomatrica or use the TE buffer otherwise the dna is stable even in high temperature
If the company is suggesting agar plate or glycerol stock is because they offer the service of extracting the DNA from your bacterial isolate prior to amplify and sequence the 16S. usually this service has an extra cost. If you can amplify your self. PCR products purified in TE has enough stability to be shipped overseas with no problem.

In my opinion, the best approach is use of FTA card from Whatman for collection and storage of blood or buccal epithelium. These samples will be preserved for a long period of time (years) in dry conditions. You can send this samples for extraction and sequencing.
Other way is Saliva DNA Collection and Preservation Devices (Norgen Biotek Corp.). I haven`t experience of use this device but it seems very interesting.

Hey Maytham. DNA is stable in TE buffer at room temperature, due to the
buffering capacity of Tris and the presence of EDTA.
