I have a sample of protiens consisting of nanofibres of 1 to 3 nanometers diameter with long length and also comprises salts like NaCl. tried with different dilution levels, but couldnot find the fibres . is there any other better way to prepare such samples? and also what is the best base on which i can coat this sample on? the same sample is done in TEM they could find few fibres,whereas the same TEM grid under SEM , I cannot find anything.is this because of the resolution limitation? or anything else?
How sure you are about the fibers? They may not be there in concentrations you think!/ Pseudofibres is one probability. The protein characteristics are one factor while the lesser or, absence of enough fibers, the coating might be difficult on the surface compatible with your preparation.

yes I was not sure about the concentration levels but I coated the sample on the Cu tape which might be drawback for appearance of nanofibers, and we tried coating on the SiO2 substrate by concentration optimization for SEM analysis . Then we could able to see them.
