There are protiens and lipid samples grown on the glass .I tried pasting a carbon tape to a side and imaging but dint work out.
Tried with various Acc.Voltage and probe current but didn't work .
Is Au or C coating the only possibility for imaging such samples?
Why didn't it work? Is the issue charging? In principle, a very low energy and a very low current, together with the right working distance should do the trick in a newer FESEM.
Why do you want to avoid coating?
Yes there is a charging issue. 2KV and 3KV I tried but no features are seen. I mean it was so noisy. After increasing the scan time as well no clarity.
Generally I use 10 mm as WD .so what is the WD u suggest .?
Its not that I want to avoid coating but we don't have a coater here soon it is going to b installed.
Can you maybe go below 1 kV? I would also use a shorter working distance, but that depends on your microscope.
