Hello. I have been reading about collagen, polycaprolactone and elastin - mixed scaffold preparation and it seems complicated, requiring special equipment. In my lab, there could be a problem with such equipment. Could you please tell me what is the absolute minimum of equipment to have in the lab if I would like to create such a mixed scaffold by myself? Or maybe you know a simple protocol to achieve this (I would prefer it to work in 96 well-plates)? I would be very grateful for your help.
Polycaprolactone basically hydrophobic in nature. so pour plate method and phase separation wont help you to provide better quality surface in your case i believe. Electrospinning is the best option. And you have to go and check different parameter starting from the preparation and the % of solution. Next about the settings, you must need voltage amplifier ,syringe with your polymer solution connected to the power pack, metal collector and the necessity protection covers from high voltages. you can find many reviews and articles which will help you in setting up this set up.
If you want to prepare porous 3d scaffolds, a very simple technique you could use is salt leaching. Other simple methods are slurry dipping for combining two polymers, of freeze drying for obtaining porous structures.
I would suggest try freeze gelation, its much more efficient than freeze drying, particulate leaching, and time saving, you just need a fridge and freezer, find out the non-solvent for solution. look up Ho et al paper on freeze gelation in 2004
