I have been reading about the possibility of measure the cell density (g/L) measuring the optical density in a spectrophotometer. What is the relation between this two parameters? Because with the spectrophotometer I can obtain absorbances but not concentrations.
Hi Noelia, Yes can do that (OD to concentration). You have many ways to do that :
1- By having standards with known concentration that you pass on spectro to get the corresponding OD, and you extrapolate to your samples.
2- By preparing the desired concentration by the reconstitution of lyophilised matrix in the diluent (water) and proceed to the OD measurements for each concentration.
3- By preparing your own standards : prepare different sets of OD and measure their corresponding dry matter content.
Through standard calibration curve you can get unknown concentration on spectrophotometer.
