Hi Anabel,

it well feasible by using Fluo-4 AM (see protocol for instance http://onlinelibrary.wiley.com/doi/10.1002/cyto.a.20974/pdf) cause Fura-2 is a ratiometric dye with double excitation in the UV range and therefore more difficult to handle in FACS analysis. I tried with Calcium crimson in microscopy but it doesn't load well, if I were you I wouldn't even try such dye.

just in order to complete the answer of Giovanni, you may use the new fluorescent calcium probe Fluo-8 AM . It is adequate for mono-excitation and mono-emission monitoring of calcium with visible light, and it is an improved version of Fluo-4. Thanks Bruno for your answer. On my lab we decided to analize ROS production of the L. mexicana parasite by flow citometry. We are using a CellROX kit from Life Tech. However, we have some troubles with the positive control of the kit (TBHP) because it does not induces ROS as it says. Do you know anything about this kit and this type of meausure on Leishmania parasites?
